Essential amino acids (valine, leucine) come from diet; non-essential (glycine, alanine) are body-synthesised; a peptide bond is an amide (–CO–NH–) link.

(a) Amino acids.

  • Essential (cannot be made by the body, must be supplied in the diet): valine, leucine (also isoleucine, lysine, phenylalanine, methionine, threonine, tryptophan, histidine, arginine).
  • Non-essential (synthesised in the body): glycine, alanine (also serine, aspartic acid, glutamic acid, etc.).

(b) Peptide linkage. When the carboxyl group (–COOH) of one amino acid condenses with the amino group (–NH₂) of another, a molecule of water is eliminated and an amide bond –CO–NH– is formed. This –CO–NH– bond joining amino-acid units in a protein is called the peptide linkage.
