A recognition sequence is the specific (usually palindromic) DNA base sequence at which a restriction enzyme binds and cuts the DNA.

Restriction endonucleases are molecular scissors used in genetic engineering. Each such enzyme does not cut DNA randomly; it inspects the length of the DNA and cuts only at a specific point.

The recognition sequence is:

  • The particular base sequence (usually a palindrome — reads the same on both strands in the 5'→3' direction) which the restriction enzyme recognises and binds to.
  • The enzyme cuts within or near this sequence.

Example: The enzyme EcoRI recognises the sequence
