A cloning vector needs: origin of replication (ori), a selectable marker, unique restriction sites for inserting DNA, small size/high copy number, and a way to screen recombinants (insertional inactivation).

A cloning vector is a DNA molecule (commonly a plasmid, bacteriophage, cosmid, or artificial chromosome) that can carry a foreign DNA fragment into a host cell and replicate there. To function well it must have the following features:

  1. Origin of replication (ori) — This is a specific sequence from which replication starts. Any piece of DNA linked to it can replicate within the host cell. This sequence also controls the copy number (the number of copies of the vector made in the cell) — a high-copy-number ori is preferred to obtain many copies of the target gene.

  2. Selectable marker — The vector must carry a marker gene (in addition to the ori) that helps identify and eliminate non-transformants and selectively permit the growth of the transformants (cells that took up the vector). Usually these are antibiotic-resistance genes (e.g. resistance to ampicillin, tetracycline, kanamycin) against which the normal E. coli cells are sensitive.

  3. Cloning / restriction sites — The vector must have single (unique) recognition sites for the commonly used restriction enzymes. If there were more than one recognition site for the same enzyme, the enzyme would cut the vector into many fragments, complicating gene cloning. The foreign DNA is ligated at such a restriction site.

  4. Small size and high copy number — A good vector should be small so that it can be easily handled and can enter the host readily, and it should multiply to a high copy number to yield many copies of the recombinant DNA.
