Cloning sites (restriction sites) are the particular sequences in a vector where a restriction enzyme cuts, allowing a piece of foreign DNA to be inserted at that exact location.

A cloning vector is a DNA molecule (such as a plasmid or bacteriophage) used to carry a foreign gene of interest into a host cell for replication/expression. To insert the foreign DNA into the vector, both the vector and the DNA fragment must be cut using the same restriction enzyme, generating matching (often complementary, "sticky") ends that can then be joined together using DNA ligase. The specific sequence within the vector where this restriction enzyme cuts and the foreign DNA is subsequently inserted is called the cloning site.
