- Transformation (CaCl2 + heat shock): Bacterial cells are first made 'competent' to take up foreign DNA by treating them with a specific concentration of divalent cations such as calcium chloride (CaCl2), which increases the efficiency with which DNA can enter through pores in the cell wall. The recombinant DNA is then incubated with these competent cells on ice, followed by briefly placing them at 42°C (heat shock) and then back on ice - this facilitates DNA entry into the bacterial cell.
- Micro-injection: In this technique, recombinant DNA is directly injected into the nucleus of an animal cell using an extremely fine glass micro-needle/micropipette, commonly used to introduce DNA into egg cells to create transgenic animals.
- Biolistics / gene gun (particle bombardment): Especially useful for plant cells with tough cell walls; DNA is coated onto high-density metal micro-particles (gold or tungsten), which are then physically shot into plant cells/tissues at high velocity using a gene gun. …
