Insertion/deletion experiments on phage genes proved codons are triplets read contiguously without gaps.

The genetic basis for this comes chiefly from the classic frameshift mutation experiments performed by Francis Crick, Sydney Brenner and colleagues on bacteriophage genes using acridine dyes (which cause insertion or deletion of a single base pair). They found that:

  • Insertion or deletion of one or two bases in a gene severely disrupted the sequence of amino acids in the protein downstream of the mutation (because the reading frame shifted), producing a non-functional protein.
  • However, insertion or deletion of three bases (or any multiple of three) restored the original reading frame after the mutated point, and the resulting protein was largely normal in the rest of its sequence. …
