  1. Name of the process: This colour-based screening method is called insertional inactivation, commonly known as blue-white selection. A gene coding for an easily-assayed enzyme (here β-galactosidase) is engineered at the site where foreign DNA is inserted; insertion of the foreign DNA disrupts (inactivates) this gene.
  2. Enzyme used: β-galactosidase, encoded by the lacZ gene. In the presence of the chromogenic substrate X-gal, bacteria with a functional (uninterrupted) lacZ gene produce β-galactosidase, which cleaves X-gal to give blue-coloured colonies (non-recombinants, i.e., the vector re-ligated without an insert). If foreign DNA has been inserted into the lacZ gene (recombinants), the gene is inactivated, no functional enzyme is made, and the colonies remain white. …
