Pyruvic acid is converted to acetyl CoA inside the mitochondrion by simultaneous loss of CO2 and oxidation — this combined process is called oxidative decarboxylation.

After glycolysis produces pyruvic acid in the cytoplasm, pyruvate enters the mitochondrial matrix for the link reaction (catalysed by the pyruvate dehydrogenase enzyme complex):

Pyruvic acid (C3) → Acetyl CoA (C2) + CO2 + NADH + H+

Two things happen together in this single step:

  1. Decarboxylation — one carbon is removed from the 3-carbon pyruvate as CO2, leaving a 2-carbon acetyl group.
  2. Oxidation — the acetyl group is oxidised (loses electrons/hydrogen, captured as NADH), and this acetyl group then combines with Coenzyme A to form acetyl CoA. …
