Site: The Krebs cycle (also called the citric acid cycle or TCA cycle) takes place in the matrix of the mitochondrion, the innermost compartment enclosed by the inner mitochondrial membrane.
The process, step by step:
- Pyruvate (from glycolysis, in the cytoplasm) is transported into the mitochondrial matrix, where it is oxidatively decarboxylated by the pyruvate dehydrogenase complex to form acetyl-CoA, releasing one CO2 and generating one NADH.
- Acetyl-CoA (2C) combines with oxaloacetate (4C) to form citrate (6C), catalysed by citrate synthase — this is the first, defining step of the cycle proper.
- Citrate is isomerised to isocitrate, then oxidatively decarboxylated to α-ketoglutarate (5C), releasing one CO2 and generating one NADH.
- α-ketoglutarate is oxidatively decarboxylated further to succinyl-CoA (4C), releasing another CO2 and generating another NADH.
- Succinyl-CoA is converted to succinate, and this step generates one molecule of GTP (readily convertible to ATP) via substrate-level phosphorylation.
- Succinate is oxidised to fumarate, generating one FADH2.
- Fumarate is hydrated to malate.
- Malate is oxidised back to oxaloacetate, generating one more NADH — and oxaloacetate is now ready to combine with a fresh acetyl-CoA and repeat the cycle. …
