Who demonstrated it: Matthew Meselson and Franklin Stahl, in a classic 1958 experiment performed on the bacterium Escherichia coli, provided the first experimental proof that DNA replicates semi-conservatively — meaning each of the two strands of the parent DNA molecule serves as a template, and each daughter DNA molecule retains one parental (old) strand and one newly synthesised strand.

Procedure in detail:

  1. E. coli cells were first grown for many generations in a medium containing ^15N (heavy nitrogen) as the sole nitrogen source, so that all the nitrogenous bases in their DNA (and hence both DNA strands) became labelled with the heavy isotope.
  2. These ^15N-labelled bacteria were then transferred to a medium containing only ^14N (normal, light nitrogen), and allowed to divide.
  3. DNA was extracted from the bacterial cultures at intervals corresponding to successive generations (after each round of replication) and subjected to caesium chloride (CsCl) density-gradient centrifugation, which separates DNA molecules by their density/weight — heavier (^15N-^15N) DNA settles lower, lighter (^14N-^14N) DNA settles higher, and hybrid DNA settles at an intermediate position.
  4. Results:
    • DNA extracted immediately after transfer (generation 0) was purely heavy (^15N/^15N band).
    • After one generation in ^14N medium, all the DNA formed a single band of intermediate (hybrid) density, meaning every DNA molecule now had one ^15N strand and one ^14N strand — ruling out conservative replication (which would have given two separate bands: pure heavy and pure light). …
