When lactose (the inducer) is present, it inactivates the repressor, allowing RNA polymerase to transcribe the lac operon's structural genes.

The lac operon consists of one regulatory gene (i) with its own promoter, and three structural genes (z, y, a) downstream of a promoter (p) and operator (o) region.

Description of the operon diagram in the presence of the inducer (lactose):

  • The same linear arrangement of boxes as in the 'absence of inducer' figure is retained: p (promoter of i) - i (regulatory gene) - p (promoter of structural genes) - o (operator) - z - y - a, with the caption 'In presence of inducer (lactose)'.
  • The regulatory gene i is transcribed as before, producing repressor mRNA, which is translated into the repressor protein.
  • Unlike the earlier (no-inducer) figure, here lactose (the inducer) is shown entering the cell and binding to the repressor protein, changing its three-dimensional conformation.
  • This inducer-bound repressor can no longer bind to the operator (o) region - so, instead of an arrow from repressor to operator blocking it, the diagram shows the repressor-inducer complex set aside, with the operator left free.
  • With the operator free, RNA polymerase binds to the promoter (p) of the structural genes and transcribes z, y, and a together as a single polycistronic mRNA. …
