  2. Reason for the disorder: Sickle cell anaemia is an autosomal recessive disorder caused by the substitution of the amino acid glutamic acid by valine at the sixth position of the beta-globin chain of haemoglobin, resulting from a point mutation (single base-pair substitution, GAG to GTG) in the gene coding for the beta-globin chain.
  • This produces an abnormal form of haemoglobin, HbS, instead of normal HbA.
