Fermentation is anaerobic glucose breakdown yielding ethanol or lactic acid plus a small amount of ATP, valuable both industrially and as an emergency energy source without oxygen.

Fermentation begins with glycolysis, in which one glucose molecule is broken down in the cytoplasm (without requiring oxygen) into two molecules of pyruvate, with a net gain of 2 ATP and 2 NADH. Under anaerobic conditions, instead of entering the mitochondria for aerobic respiration, this pyruvate is further converted:

  • In alcoholic fermentation (e.g. in yeast), pyruvate is converted to acetaldehyde and then to ethanol, releasing CO2, while regenerating NAD+ needed to keep glycolysis running.
  • In lactic acid fermentation (e.g. in Lactobacillus bacteria, and in human muscle cells during intense exercise when oxygen supply is insufficient), pyruvate is directly converted to lactic acid, again regenerating NAD+. In both cases, the overall ATP yield is small - just the 2 ATP produced during glycolysis - since the pyruvate is not fully oxidised as it would be in aerobic respiration.
