The valine codon involved in sickle-cell haemoglobin is GUG, arising from a single-base substitution (A→U) in the glutamic acid codon GAG.

Haemoglobin is built from two α and two β globin polypeptide chains. Normal adult haemoglobin (HbA) has glutamic acid (Glu) as the sixth amino acid of the β-chain, coded by the mRNA codon GAG (or GAA).

In sickle-cell anaemia, a point mutation in the β-globin gene changes a single DNA base pair, which changes the mRNA codon from GAG to GUG. Since GUG codes for valine, glutamic acid (a polar, negatively charged amino acid) is replaced by valine (a non-polar, hydrophobic amino acid) at the sixth position of the β-chain.
