A palindromic DNA sequence reads identically on both strands when each is read 5'→3', a property exploited by restriction enzymes to recognise their cut sites.

Palindromic nucleotide sequence: In the context of DNA, a sequence of base pairs is said to be palindromic if it reads the same when read in the 5'→3' direction on one strand as it does when the complementary strand is also read in the 5'→3' direction. In other words, the sequence has a two-fold (dyad) symmetry, such that:

5'—G A A T T C—3'

3'—C T T A A G—5'

Reading the top strand 5'→3' gives GAATTC; reading the bottom strand 5'→3' (i.e., from right to left as drawn) also gives GAATTC — this sequence is therefore palindromic.
