Common methods for introducing a recombinant/foreign DNA into a host cell include:
- Chemical (CaCl2) transformation: treating recipient bacterial cells with a specific concentration of a divalent cation such as Ca2+, which increases the efficiency of DNA entry through the cell wall pores, followed by briefly incubating the cells with recombinant DNA on ice and then giving a heat shock (42°C for ~90 seconds) followed by cooling on ice — this makes the bacterial cell 'competent' to take up the foreign DNA.
