The lac operon is Jacob and Monod's classic inducible operon in E. coli: a repressor blocks the operator by default, and lactose relieves this block to switch on the structural genes needed to metabolise it.

A schematic/diagrammatic representation cannot be drawn as an image here, but its layout and function can be described precisely, left to right along the DNA:

Components (in order on the DNA):

  1. Regulatory gene (i): Located upstream, it is constitutively transcribed and continuously produces the lac repressor protein.
  2. Promoter (p): The site where RNA polymerase binds to begin transcription of the structural genes.
  3. Operator (o): Lies next to the promoter; the repressor protein binds here when active, physically blocking RNA polymerase from transcribing the structural genes.
  4. Structural genes - z, y, a:
    • z gene codes for beta-galactosidase (hydrolyses lactose into glucose + galactose).
    • y gene codes for permease (transports lactose into the cell).
    • a gene codes for transacetylase. All three are transcribed together as a single polycistronic mRNA.

Regulation (schematic logic):

  • In the absence of lactose: The repressor (from gene i) binds tightly to the operator (o), physically blocking RNA polymerase at the promoter from transcribing z, y, a. The operon is OFF. …
