Water splitting (photolysis) takes place on the lumenal side of the thylakoid membrane at PS II's oxygen-evolving complex, and it is the source of both the electrons that keep the light reaction running and the O2 released in photosynthesis.

Site: Photolysis of water occurs within the thylakoid lumen, at a manganese-containing protein complex called the oxygen-evolving complex (OEC), which is physically associated with Photosystem II (specifically its reaction centre, P680).

Reaction: 2H2O -> 4H+ + 4e- + O2

Significance:

  1. It supplies electrons that replace those lost from the PS II reaction centre (P680) when it is excited by light and donates electrons down the electron transport chain toward PS I. Without this replenishment, PS II could not continue functioning.
