A test cross with a homozygous recessive reveals genotype: monohybrid gives 1:1, dihybrid gives 1:1:1:1.

Test cross — concept. A test cross is a cross of an individual showing the dominant phenotype (whose genotype is unknown) with a homozygous recessive individual. The phenotypic ratio of the offspring reveals whether the dominant parent is homozygous or heterozygous, so it is used to determine an unknown genotype.

Monohybrid test cross (one character). Cross the F₁ tall Tt with dwarf tt:

  • Gametes of Tt: T, t; gamete of tt: t.
  • Offspring: Tt (tall) and tt (dwarf) in a 1 : 1 ratio.

Since tall and dwarf appear in equal numbers, the tested parent must be heterozygous (Tt). (If it were TT, all offspring would be tall.)

Dihybrid test cross (two characters). Cross F₁ TtRr (tall, round) with recessive ttrr (dwarf, wrinkled):

  • Gametes of TtRr: TR, Tr, tR, tr; gamete of ttrr: tr. …
