Recombinant DNA technology involves isolation of DNA, cutting with restriction enzymes, ligation into a vector, transformation into a host, and selection/screening of recombinants.

The main processes/steps of Recombinant DNA (rDNA) technology are: (1) Isolation of genetic material (DNA) from the source organism/cell. (2) Cutting (fragmentation) of DNA at specific sites using restriction endonuclease enzymes (molecular scissors), generating a gene of interest and a cut vector DNA with complementary ('sticky') ends. (3) Ligation — joining the gene of interest with the cut vector DNA using the enzyme DNA ligase, producing a recombinant DNA molecule. (4) Introducing/transferring the recombinant DNA into a host cell (bacterium, yeast, plant or animal cell) — a process called transformation, done via methods such as heat-shock/CaCl2 treatment, micro-injection, gene gun (biolistics), or use of a disarmed pathogen vector like Agrobacterium. (5) Obtaining the foreign gene product — culturing the transformed host cells so they multiply and express the i …
