Sickle cell anaemia offers one of the clearest windows into how a tiny molecular error can cascade into a life-altering disease. The condition stems from a point mutation — a change in just one nucleotide in the DNA sequence of the gene coding for the β-globin chain of haemoglobin. This single-letter typo in the genetic code has profound consequences.

In normal haemoglobin (HbA), the sixth amino acid in the β-globin chain is glutamic acid, a hydrophilic (water-loving) amino acid. The codon that specifies glutamic acid is GAA or GAG in mRNA. In sickle cell anaemia, a point mutation changes the middle nucleotide of this codon: on the coding (sense) strand, an adenine (A) is replaced by a thymine (T), so the mRNA codon GAG becomes GUG. This new codon now codes for valine, a hydrophobic (water-repelling) amino acid.

That single substitution — glutamic acid to valine — alters the entire structure of haemoglobin. Valine's hydrophobic nature causes the mutant haemoglobin molecules (HbS) to stick together under low oxygen conditions, forming long, rigid polymers.
