Hofmann bromamide degradation shortens the chain by one carbon and gives pure primary amines with retention of configuration — both are its key significances.

In the Hofmann bromamide degradation reaction, a primary amide reacts with bromine in the presence of concentrated NaOH/KOH to give a primary amine with one carbon atom fewer than the parent amide:

RCONH2+Br2+4NaOH→RNH2+Na2CO3+2NaBr+2H2ORCONH_2 + Br_2 + 4NaOH \rightarrow RNH_2 + Na_2CO_3 + 2NaBr + 2H_2O

Significance 1 — Chain shortening / clean synthesis of 1° amines: Because the product amine has exactly one carbon less than the starting amide, this reaction is a reliable way to descend a homologous series by one carbon. Unlike ammonolysis of alkyl halides (which often gives a mixture of 1°, 2°, 3° amines and quaternary salts due to further alkylation), Hofmann degradation gives ONLY the primary amine, cleanly and in good yield.
