(b) Nucleosome:
- In the eukaryotic nucleus, the long, negatively charged DNA is packaged with positively charged basic proteins called histones.
- A set of eight histone molecules (two molecules each of H2A, H2B, H3 and H4) organise to form a unit called the histone octamer.
- The negatively charged DNA is wrapped around this positively charged histone octamer to form a structure called a nucleosome. A typical nucleosome contains about 200 base pairs of DNA.
- Nucleosomes are the repeating units of chromatin; under the electron microscope, chromatin appears as 'beads-on-a-string', where the beads are the nucleosomes. This packaging shortens the DNA and is the first level of DNA condensation.
(c) Transfer RNA (tRNA):
- tRNA (earlier called soluble RNA, sRNA) is an adapter molecule proposed by Francis Crick that acts as a link between the codon on mRNA and the amino acid.
- It reads the genetic code: it has an anticodon loop that has bases complementary to the codon on mRNA, and an amino-acid acceptor end (the 3' —CCA end) to which the specific amino acid is attached (charged tRNA/aminoacyl-tRNA).
- Its secondary structure is depicted like a clover-leaf, but in three dimensions it looks like an inverted 'L'.
- There is a specific tRNA for each amino acid, and a special initiator tRNA (carrying methionine) that starts translation. There are no tRNAs for the stop codons. …
