Two main uses of a potentiometer: comparing/measuring the EMF (and internal resistance) of cells, and measuring an unknown potential difference (or calibrating meters).

A potentiometer works on the principle that the potential drop across a uniform wire carrying a steady current is directly proportional to the length of the wire (V∝lV \propto l). Because at the balance (null) point it draws no current from the cell being tested, it acts as an ideal voltmeter of infinite resistance and gives the true EMF.

Use 1 — Comparison of EMFs of two cells: By finding balancing lengths l1l_1 and l2l_2 for the two cells, E1E2=l1l2\dfrac{E_1}{E_2} = \dfrac{l_1}{l_2}. The same set-up (with a cell in an auxiliary circuit) measures the internal resistance of a cell.
