Melvin Calvin traced the path of radioactively labelled carbon dioxide through photosynthesising algae, which allowed him to identify the first product of CO2 fixation and reconstruct the entire cyclic pathway that now bears his name.

In the period just after World War II, among several efforts to find beneficial applications for radioisotope technology, Melvin Calvin's work on photosynthesis stood out. He used algae supplied with carbon dioxide labelled with the radioactive isotope 14C, allowing him to follow, step by step, where the carbon from that CO2 ended up as photosynthesis proceeded.

Through this tracing, Calvin discovered that the very first stable product formed when CO2 is fixed was not, as some had guessed, built directly around a small 2-carbon acceptor, but was in fact a 3-carbon organic acid — later identified specifically as 3-phosphoglyceric acid, or PGA. This was, in itself, an important finding, since it also led to the identification of the actual CO2 acceptor as the unexpected 5-carbon sugar ribulose 1,5-bisphosphate (RuBP), rather than the 2-carbon molecule scientists had long assumed. …
