a) Importance of Agarose gel:

Agarose (a natural polymer extracted from seaweed) is cast into a gel with a mesh-like network of pores. When an electric current is applied, DNA fragments (negatively charged due to the phosphate backbone) migrate towards the positive electrode (anode) through this mesh. Smaller fragments move faster and travel farther through the pores, while larger fragments move more slowly — separating the mixture into distinct bands according to their size (molecular weight). This is the fundamental separating medium of gel electrophoresis.
