Gel electrophoresis separates charged molecules (usually DNA fragments) by size by driving them through a gel with an electric current.

Gel electrophoresis is a laboratory technique used to separate DNA (or RNA/protein) fragments on the basis of their size (length).

How it works:

  • DNA is negatively charged (because of its phosphate groups), so when placed in an electric field it moves towards the positive electrode (anode).
  • The fragments are made to move through a matrix of agarose gel (a natural polymer from sea weeds), which acts like a molecular sieve.
  • Smaller fragments move faster and farther; larger fragments move slowly and stay near the wells. Thus fragments get separated according to size. …
