By radioactively labelling phage DNA and protein separately, Hershey and Chase showed only DNA enters bacteria during infection, confirming DNA as the genetic material.

The Hershey-Chase Experiment (1952):

  1. Alfred Hershey and Martha Chase worked with bacteriophage T2, a virus that infects E. coli and consists only of a protein coat and DNA.
  2. They grew one batch of phages in a medium containing radioactive phosphorus (³²P), which is incorporated into DNA (since DNA contains phosphate but protein does not), labelling the phage DNA.
  3. They grew a second batch of phages in a medium containing radioactive sulfur (³⁵S), which is incorporated into proteins (since sulfur-containing amino acids, cysteine and methionine, occur in protein but not DNA), labelling the phage protein coat.
  4. Each labelled batch was allowed to infect separate cultures of E. coli.
  5. After infection, the cultures were agitated in a blender to shear off the empty phage coats attached to the bacterial surface, and then centrifuged to separate the heavier bacterial cells (pellet) from the lighter phage coats (supernatant). …
