Hershey-Chase (1952) settled the DNA-vs-protein debate by radioactively labelling phage DNA and protein separately and tracking which one entered the bacterium during infection — only the DNA did.

Reason for the experiments: By the early 1950s it was known that bacteriophages consist of only DNA and a protein coat, but it was not yet clear which of the two — DNA or protein — actually carried the genetic information passed from the virus into the host bacterium during infection. Hershey and Chase designed their experiment specifically to determine, conclusively, which of these two molecules is the genetic material.
