Isolating genetic material involves breaking open the cell, removing contaminating RNA and protein, and then precipitating the purified DNA using chilled ethanol.

Since DNA is present inside the cell (and nucleus, in eukaryotes), along with many other macromolecules such as RNA and proteins, isolating pure DNA requires the following steps:

  1. Breaking open the cell (lysis): appropriate enzymes are used depending on the cell type — lysozyme to break bacterial cell walls, cellulase for plant cell walls, and chitinase for fungal cell walls — to release the cell's contents, including the DNA, into solution along with other biomolecules.
  2. Removing other macromolecules: the crude extract is treated with ribonuclease (RNase) to degrade and remove RNA, and with protease to remove/degrade proteins that are bound to the DNA (such as histones), so that mainly purified DNA remains in solution. …
