An enzyme's catalytic cycle is the repeating sequence of substrate binding, transformation, product release, and enzyme regeneration that allows one enzyme molecule to catalyse a reaction over and over.

An enzyme catalyses a reaction through the following steps, which together form one catalytic cycle:

  1. Substrate binds to the active site of the enzyme, fitting in a specific orientation (as per the lock-and-key or, more accurately, induced-fit model).
  2. The active site changes shape slightly to more precisely fit around the substrate once it is bound, forming an enzyme-substrate (ES) complex. This 'induced fit' also facilitates the reaction.
  3. The active site of the enzyme, now in close proximity to the substrate, acts on it to break/form the relevant chemical bonds, converting the substrate into a product, or, in the case of a reaction involving two substrates, aligning them so they can react together. This lowers the activation energy needed for the reaction.
  4. The enzyme releases the product(s) of the reaction, and the free enzyme is now ready to bind to another molecule of substrate. …
