Chromatography separates mixture components based on their differing affinities for a stationary phase versus a mobile phase; common types are column chromatography, paper chromatography, and thin-layer chromatography (TLC).

Chromatography is based on the principle that different substances in a mixture travel at different rates through/over a stationary phase (a solid or a liquid supported on a solid) when carried by a moving mobile phase (a liquid or a gas), because each component is adsorbed onto (or dissolves in) the stationary phase to a different extent, or is carried along by the mobile phase to a different extent.

Main types:

  1. Adsorption chromatography: based on differential adsorption of components on an adsorbent stationary phase (e.g., alumina, silica).
    • Column chromatography: the mixture is placed at the top of a column packed with an adsorbent; different components are eluted (washed out) at different rates by a solvent, and separately collected.
    • Thin-Layer Chromatography (TLC): a thin layer of adsorbent is coated on a glass/plastic plate; the mixture spot moves up the plate as solvent rises by capillary action, separating into distinct spots. …
