PCR is a laboratory technique to make millions of copies of a specific DNA (gene) sequence using repeated cycles of denaturation, primer annealing and extension.

What is PCR?

Polymerase Chain Reaction (PCR) is a technique used to amplify (make multiple copies of) a gene/DNA segment of interest in vitro, using a pair of short chemically synthesised oligonucleotide primers complementary to the two ends of the target sequence, a heat-stable DNA polymerase (commonly Taq polymerase, isolated from the thermophilic bacterium Thermus aquaticus), and repeated cycles of a thermal reaction.

The three steps of one PCR cycle:

  1. Denaturation — the double-stranded DNA template is heated (~94–96 °C) to separate it into two single strands.
  2. Annealing (Primer annealing) — the reaction is cooled, allowing the two primers to bind (hybridise) to their complementary sequences on the single-stranded template. …
