Any candidate genetic material must satisfy four criteria: accurate replication, stability, capacity to mutate, and expression as phenotype.

For any molecule to qualify as the genetic material of a living organism, it must fulfil the following properties/criteria:

  1. Replication — it must be able to replicate itself accurately, so that genetic information can be faithfully copied and passed on from parent to offspring across generations.

  2. Chemical and structural stability — it must be chemically and structurally stable enough that the genetic information encoded in it is preserved reliably, largely unaltered, through the various stages of the life cycle and environmental conditions. DNA is more stable than RNA because deoxyribose lacks the reactive 2'-OH group present in ribose (making RNA more easily degraded), and because DNA uses thymine (5-methyl uracil) instead of uracil — this allows any cytosine that spontaneously deaminates to uracil (a mutational change) to be recognised as 'foreign' and repaired, a correction mechanism not available if uracil were a normal DNA base.

  3. Capacity to mutate — it must provide scope for slow but steady, heritable changes (mutations), since mutations are the raw material that permits variation, upon which natural selection acts, and hence is essential for evolution.
