DNA fingerprinting identifies individuals by detecting unique patterns in repetitive DNA sequences, through a multi-step process of digestion, separation, blotting and probing — widely applied in forensics and paternity testing.

  1. What is DNA fingerprinting? DNA fingerprinting is a technique that identifies differences (polymorphisms) in specific regions of an individual's DNA — particularly in repetitive DNA sequences called VNTRs (Variable Number of Tandem Repeats), which vary enormously in length between individuals (except identical twins) — producing a pattern of DNA bands unique to each person, analogous to a physical fingerprint.
  2. Steps in DNA fingerprinting:

  2. Restriction digestion — cutting the DNA into fragments using restriction endonuclease enzymes.
  3. Separation by gel electrophoresis — sorting the DNA fragments by size, which migrate different distances through the gel.
  4. Southern blotting — transferring (blotting) the separated DNA fragments from the gel onto a synthetic membrane (e.g., nitrocellulose or nylon).
  5. Hybridisation — the membrane is probed with a labelled (radioactive) VNTR probe that binds specifically to the complementary repetitive sequences.
  6. Detection (autoradiography) — an X-ray film is exposed to the membrane, revealing a pattern of dark bands corresponding to the hybridised, labelled fragments — this banding pattern is the 'DNA fingerprint'.
