EcoRI recognises the palindromic sequence 5'-GAATTC-3' / 3'-CTTAAG-5' and cleaves between G and A, generating complementary sticky (cohesive) ends.

Concept. Restriction endonucleases are the 'molecular scissors' of recombinant DNA technology. Each enzyme recognises a specific short palindromic sequence and cuts at a fixed position within or near it.

The EcoRI site. A palindrome reads the same 5'→3' on both strands. EcoRI recognises six base pairs and cuts each strand between the guanine (G) and adenine (A):

5′−G↓A A T T C−3′5'-\text{G} \downarrow \text{A A T T C}-3'

3′−C T T A A↑G−5′3'-\text{C T T A A} \uparrow \text{G}-5'
