PCR amplifies a target DNA segment in vitro; the heat-stable Taq DNA polymerase used comes from Thermus aquaticus.

Use of Polymerase Chain Reaction (PCR):

  • PCR is a technique to synthesise multiple copies of a specific gene/DNA segment (amplification) outside the living cell, in vitro.
  • Using two primers and DNA polymerase, the amount of target DNA can be increased by nearly a billion times through repeated cycles of denaturation, annealing and extension.
  • Amplified DNA is used in genetic engineering, molecular diagnosis of diseases, detection of pathogens/mutations, DNA fingerprinting and gene cloning.
