A retrovirus with RNA genome uses reverse transcriptase to make DNA from its RNA; this DNA integrates into host DNA and directs production of new viral RNA and proteins, forming new viruses.

Schematic representation of retrovirus (HIV) replication:

  1. Entry – The retrovirus (containing single-stranded RNA and the enzyme reverse transcriptase) enters the host cell (e.g., a helper T-lymphocyte / macrophage) after binding to it.

  2. Reverse transcription – Inside the host cell, the enzyme reverse transcriptase synthesises a complementary DNA (cDNA) copy using the viral RNA as a template (RNA → DNA). This is the reverse of the usual central dogma.

  3. Integration – The newly formed viral DNA gets incorporated into the host cell's genome (DNA) and is called a provirus.

  4. Transcription – The integrated viral DNA directs the host machinery to produce new viral RNA (genomic RNA) and mRNA.
