The mesophyll -- the photosynthetic ground tissue between the two epidermal layers of a leaf -- is differentiated into a compact palisade layer near the upper surface and a loosely packed, air-space-rich spongy layer near the lower surface, close to the stomata.

In a dorsiventral (dicot) leaf like the one in the figure, the tissue lying between the adaxial (upper) epidermis and the abaxial (lower) epidermis, and surrounding the vascular bundle (with its xylem, phloem, and bundle sheath), is called the mesophyll. From the figure, three features of the mesophyll cells can be noted:

  1. The mesophyll is differentiated into two distinct regions: an upper palisade layer of cells lying just beneath the adaxial epidermis, and a lower spongy layer of cells lying just above the abaxial epidermis (near the stomata).

  2. The palisade mesophyll cells are elongated, columnar (rod-shaped) cells, arranged vertically and packed compactly/closely together with little intercellular space between them. Because they contain numerous chloroplasts, they are the main site of photosynthesis in the leaf.
