To get a virus-free plant from an infected one, tissue culture uses the shoot-tip (apical meristem) as the explant, because the actively-dividing meristematic tissue generally stays free of the virus even when the rest of the plant is infected.

  2. Reason: The cells of the shoot apical meristem are meristematic — they divide extremely rapidly. A virus present in the infected plant spreads through the plant body via the vascular tissue (phloem) at a slower rate than the meristematic cells proliferate, and the meristematic dome itself lacks a well-developed vascular supply. As a result, the growing tip of an infected plant usually remains free of the virus even though the mature tissues below it are infected.
