Lysosomes and vacuoles are both endomembrane structures, but lysosomes are enzyme-filled digestive vesicles that break down macromolecules, while vacuoles are single-membrane storage compartments that hold water, sap and wastes and can also carry out osmoregulation.

Several membrane-bound organelles inside a eukaryotic cell are grouped together as the endomembrane system because their activities are coordinated with one another. Among its members are both the lysosomes and the vacuoles. So they share a family membership — yet a comment on them must bring out how differently they behave, because having a membrane is not the same as having the same function.

Lysosomes are tiny, membrane-bound vesicular structures. They are produced through the packaging activity of the Golgi apparatus, originating as vesicles pinched off from it. What makes them distinctive is their contents: when isolated and studied they are found to be extremely rich in almost every type of hydrolytic enzyme — lipases that act on lipids, proteases that act on proteins, and carbohydrases that act on carbohydrates. These enzymes are optimally active under acidic conditions. Through this collection of enzymes, lysosomes are able to digest all the major classes of biological molecules: carbohydrates, proteins, lipids and nucleic acids. Their role, in a word, is digestion.

Vacuoles are membrane-bound spaces within the cytoplasm. Rather than being solid, a vacuole is an enclosed pocket holding a variety of contents — water, sap, excretory products and other substances the cell does not need.
