Aminoacylation activates a tRNA by attaching its cognate amino acid (using ATP), a two-step process essential before that amino acid can be added to a growing polypeptide during translation.

Part (i) — Aminoacylation of tRNA:

Aminoacylation, or "charging" of tRNA, is the process by which a specific amino acid is covalently attached to its corresponding tRNA, catalysed by a specific enzyme, aminoacyl-tRNA synthetase (a different synthetase exists for each of the 20 amino acids, ensuring high fidelity). It occurs in two steps:

  1. Activation: amino acid + ATP → aminoacyl-adenylate (aminoacyl-AMP) + PPi, catalysed by the aminoacyl-tRNA synthetase; this energises the amino acid.
  2. Transfer: the activated amino acid is transferred from aminoacyl-AMP onto the 3′-CCA end of its specific tRNA, forming aminoacyl-tRNA (charged tRNA) + AMP.

This charged tRNA is then delivered to the ribosome, where its anticodon pairs with the corresponding mRNA codon, allowing the amino acid to be added to the growing polypeptide chain by peptide-bond formation. Without aminoacylation, tRNA cannot deliver amino acids during translation.
