Gene therapy attempts to correct a genetic disorder at its root by delivering a working copy of the missing/defective gene into a patient's cells, illustrated by the treatment of ADA deficiency.

Gene therapy is a set of approaches that attempt to correct a genetic defect diagnosed in a child or embryo, by delivering a normal, functional gene into the individual's cells or tissues to make up for the effect of a faulty, non-functional gene, thereby treating a genetic disorder at its molecular origin.

Example: Adenosine Deaminase (ADA) deficiency. ADA deficiency is caused by the absence or deletion of the gene for the enzyme adenosine deaminase, which is essential for proper functioning of the immune system (its deficiency causes Severe Combined Immuno-Deficiency, SCID). In the first clinical gene-therapy trial, lymphocytes were removed from the patient's blood, and a normal, functional copy of the ADA gene was introduced into these lymphocytes using a retroviral vector; these genetically corrected cells were then returned to the patient's body.
