Within the chloroplast, ATP/NADPH synthesis and water photolysis happen on the thylakoid membrane system, while CO2 fixation, sugar synthesis and starch synthesis all take place in the surrounding stroma.

The chloroplast shows a clear division of labour between its membrane system and its stroma, and each of the processes in this question falls cleanly into one location or the other.

a. Synthesis of ATP and NADPH: This happens on the membrane system of the chloroplast — the grana and the stroma lamellae — since this membrane system is responsible for trapping light energy and, through electron transport and the chemiosmotic mechanism, synthesising both ATP and NADPH.

b. Photolysis of water: This occurs specifically at the water splitting complex, which is associated with Photosystem II and is located on the inner side of the thylakoid membrane. Because of this location, the protons and oxygen released by splitting water end up in the thylakoid lumen rather than in the stroma.

c. Fixation of CO2: This is the carboxylation step of the Calvin cycle, and it takes place in the stroma, the fluid matrix surrounding the thylakoid membranes, where the enzyme RuBisCO catalyses the addition of CO2 to RuBP. …
