Transcription copies one strand of DNA into RNA via RNA polymerase, in initiation, elongation and termination steps.

Transcription is the process by which the genetic information encoded in a segment (gene) of one strand of DNA — the template or antisense strand — is copied into a complementary single strand of RNA. It is catalysed by RNA polymerase and occurs in three stages:

  1. Initiation: RNA polymerase, with the help of transcription (sigma) factors, recognises and binds to the promoter sequence upstream of the gene, unwinding the DNA to expose the template strand and forming the transcription "bubble".
  2. Elongation: RNA polymerase moves along the template strand in the 3'→5' direction, synthesising the RNA transcript in the 5'→3' direction by adding ribonucleotides complementary to the template (using uracil in place of thymine). Only a short stretch of DNA is unwound at any time, and the DNA re-winds behind the moving enzyme. …
