The carbylamine reaction converts a primary amine + CHCl3CHCl_3/KOH into a foul-smelling isocyanide, and is used as a diagnostic test unique to primary amines.

When a primary amine (either aliphatic, RNH2RNH_2, or aromatic, ArNH2ArNH_2) is heated with chloroform (CHCl3CHCl_3) and alcoholic potassium hydroxide (KOH), it is converted into an alkyl/aryl isocyanide (carbylamine), which has an extremely unpleasant, offensive odour:

RNH2+CHCl3+3KOH→ΔRNC+3KCl+3H2ORNH_2 + CHCl_3 + 3KOH \xrightarrow{\Delta} RNC + 3KCl + 3H_2O

C6H5NH2+CHCl3+3KOH→ΔC6H5NC+3KCl+3H2OC_6H_5NH_2 + CHCl_3 + 3KOH \xrightarrow{\Delta} C_6H_5NC + 3KCl + 3H_2O

Mechanistically, the alcoholic KOH first generates dichlorocarbene (:CCl2:CCl_2) from chloroform, which is then attacked by the lone pair of nitrogen on the primary amine; subsequent loss of two more HCl equivalents (with base) gives the isocyanide.
