Step 1. A total RBC or WBC count, obtained by diluting blood with the appropriate fluid (Hayem's fluid for RBCs, Turk's solution for WBCs) and counting under the microscope with a haemocytometer, is reported as the number of cells per cubic millimetre (mm3) of blood.

The accepted normal reference range for total red blood cells in a healthy adult is approximately 4.5 to 5.5 million cells per mm3 of blood - red cells being by far the most numerous formed element in blood, which is what makes the haemocytometer's RBC grid finer (25 tertiary squares) than its WBC grid.
