The C3 and C4 labels refer purely to the number of carbon atoms in the very first stable compound formed when a plant fixes CO2 — three carbons for C3 plants, four for C4 plants.

Early researchers studying CO2 fixation wanted to know what the first product was when CO2 is fixed by a plant. Melvin Calvin's work with radioactive carbon in algae showed that in many plants, the first CO2-fixation product was a 3-carbon organic acid, later identified as 3-phosphoglyceric acid (PGA). Because this molecule has three carbon atoms, plants that follow this route are said to use the C3 pathway.

Scientists later discovered that not all plants form PGA first. In another group of plants — those adapted to dry tropical conditions — the first stable product of CO2 fixation was found to be a different organic acid, this time with four carbon atoms, identified as oxaloacetic acid (OAA). Because this first product carries four carbons, these plants are said to use the C4 pathway. …
