Pre-rDNA insulin came from animal pancreases and could trigger immune reactions; genetically engineered insulin is made by producing the human A and B chains separately in E. coli and chemically joining them.

Before rDNA technology: Insulin used to treat diabetics was extracted and purified from the pancreases of slaughtered cattle and pigs. Although this animal insulin (bovine/porcine insulin) is effective at controlling blood glucose, its amino acid sequence differs slightly from human insulin, so it was foreign to the human body and could trigger allergic reactions or an immune response in some patients over prolonged use.

Steps in producing genetically engineered (human) insulin:

Mature, functional human insulin consists of two short polypeptide chains -- Chain A (21 amino acids) and Chain B (30 amino acids) -- linked together by disulfide bonds. (In the body it is initially synthesised as a single polypeptide, proinsulin, which folds and is then cleaved to remove the connecting C-peptide, leaving the two chains joined by disulfide bridges.)

  1. The DNA sequences coding for the human insulin A chain and B chain were chemically synthesised.
  2. These two DNA sequences were separately inserted into plasmid vectors and introduced into two different E. coli cultures.
