Q.Insulin in the human body is secreted by pancreas as prohormone/proinsulin. The schematic polypeptide structure of proinsulin is given below. The proinsulin needs to undergo processing before it becomes functional in the body. Observe figure and answer the questions that follows.

(a) State the changes the Proinsulin undergoes at the time of processing to become functional.
(b) How are the two Polypeptide of a functional insulin chemical held together?

Proinsulin, which has three polypeptide segments (A, B and a connecting C-peptide), is converted to mature, functional insulin by removal of the C-peptide during processing; the remaining A and B chains stay linked to each other through disulfide (S–S) bonds.

Figure: A schematic of the proinsulin polypeptide drawn as one long looping chain labelled 'Proinsulin', forming a large open loop at top labelled 'C' (the connecting/C-peptide region), which comes down into two parallel horizontal chain segments below labelled 'A' (upper segment) and 'B' (lower segment); two 'S-S' disulfide-bond labels mark cross-links between the A and B segments.

Insulin used to treat diabetes (e.g. Humulin, produced by recombinant DNA technology in E. coli by Eli Lilly) is first synthesised as a single-chain precursor called proinsulin, consisting of the A chain, the B chain, and a connecting C-peptide joining them.
